1 Abstract

There is provided a process of preparing oligodeoxyribonucleotides which comprises incorporating ribonucleotide units at specific locations in deoxyribonucleotide chains, to provide predetermined cleavage sites which allow ease of chain cleavage. In one embodiment, a ribo unit is used as the a-unit to couple directly to a polymer support in synthesis, to increase the polymer-nucleoside initial loading. In another embodiment ribo units are used near the m-end of the chain, followed by a small terminal nucleotide sequence which can be used to separate the product from unwanted products by means of an affinity column. The polynucleotide chain can be treated with base or RNAse, to remove the ribo units therefrom after synthesis and purification.
