Description
Enables cysteine-type endopeptidase activity and protein homodimerization activity. Involved in several processes, including nervous system development; programmed cell death involved in cell development; and protein processing. Located in cytoplasm; nucleus; and plasma membrane. Part of apoptosome. Is expressed in several structures, including clypeo-labral primordium; cystic bulge; germline cyst; gut section; and salivary gland. Used to study testicular cancer. Human ortholog(s) of this gene implicated in autosomal recessive intellectual developmental disorder. Orthologous to human CASP2 (caspase 2).

2.1 Protein Function

Involved in the activation cascade of caspases responsible for apoptosis execution (PMID: 10200258, PMID: 10675329, PMID: 10984473). Effector of steroid-mediated apoptosis during insect metamorphosis (PMID: 10984473). Overexpression promotes programmed cell death (PMID: 10200258, PMID: 10675329, PMID: 10984473). Interaction with Diap1 is required to suppress Dronc-mediated cell death; via Diap1-mediated ubiquitination of Dronc (PMID: 10675329, PMID: 14517550). Rate-limiting caspase in rpr, grim and hid death pathway (PMID: 10984473, PMID: 14517550). Recruited to the Dark apoptosome, an adapter protein complex that mediates activation of the caspase cascade in programmed cell death initiated by the intrinsic apoptosis pathway (PMID: 21220123, PMID: 25644603). Association with the Dark apoptosome stimulates autocatalytic cleavage and activation of Dronc, promoting Dronc-mediated cleavage of downstream effector caspases such as Drice (PMID: 21220123, PMID: 25644603).
