Description:
This assay was used to identify small molecules capable of enhancing the survival of dissociated human embryonic stem cells.
Protocol
5000 dissociated human embryonic stem cells were plated per well of a 384 well plate and incubated with small molecules for 4 days. At the end of the assay, cells were stained for a marker of pluripotent cells and the number of stem cells was calculated using high content image analysis software.
Comment
Activity Score was determined by normalizing the number of OCT4 positive hESCs detected (Positive W2) to a 0-100 scale. Activity scores for active compounds are 40-100, activity scores of inactive compounds are 0-40.
